The aim of the study was to evaluate the immune response reflecting function of Th1 and Th2- lymphocytes and cytokine profile (IFN- ?, IL- 2, IL - 4, IL -6, IL -10 concentration in the blood) in albino rats with chronic intoxication with ethanol (E ). Experiments were performed on male and female albino rats weighing 180-240 g were administered per os ethanol 40 % aqueous solution of 0,2 DL50 per dose for 30 days (DL50 ethanol was 12,3+1,3 g / kg). Indicators of immunity was assessed after 30 days by standard methods in immunology . Immunization of
sheep erythrocytes (2 o 108of evaluation function e for Th1- cells ( for IgM production and delayed type hypersensitivity reactions ) and Th2- lymphocytes ( product IgG). Cytokine concentration in the blood was determined by enzyme-linked immunosorbent assay (ELISA) after 30 days. Immune system parameters characterizing Th1- lymphocyte function , the action of E on average decreased by
3.0 times , and the indicators associated with the function of Th2- cells - in 2,1 times . The content of IFN -? and IL -4 was reduced respectively by 3.8 and 2.8 -fold (p <0,05). Comparison IFN-?/IL-4 influenced E was 5.6 (the control - 7.5), which indicates a greater reduction function Th1- lymphocytes compared to the Th2- cells. Concentrations of IL- 2, IL -6 and IL -10 after chronic intoxication E decreased respectively 3.1, 2.05 and 1,87 (p <0,05). Reducing the
concentration of IL -2 indicates the suppression of its production by T-lymphocytes , natural killer cells , reduction of proliferation of T and B cells . Reduction in blood IL -6 characterized by the inhibitory effect of E Th0- and Th2- cells , fibroblasts , macrophages and B cells , and IL -10 - by macrophages, dendritic cells of lymphoid , Th0- and Th2- cells and B - cell dependent macrophage proliferation of Th1- cells. Thus , chronic intoxication E significantly reduces the concentration of blood cytokine IFN -?, IL -2 , and to a lesser degree - the IL -4 , IL- 6, IL -10 causes a greater loss Th1- cells compared to the Th2- lymphocytes and IFN-?/IL-4 reduces the ratio compared with the control.
